Özet
Aim In this study, we evaluated the capacity of Gossypin in H 2 O 2induced oxidative stress and wound healing model in mouse L929 fibroblast cells. Material and Method In vitro wound healing activity, cell viability and cytotoxicity indexes of Gossypin (5-200 µg/mL) and H 2 O 2 (0.3-0.9 mM) were evaluated in the mouse fibroblast cell line L929 using the MTT assay. Migration test was performed as an in vitro wound healing test model. The concentration of H 2 O 2 (0.5 mM) was used to evaluate the antioxidant activity. SOD activity and MDA level were determined. We also analyzed IL-1β and TNF-α mRNA expression levels in L929 cells using real-time PCR to assess whether Gossypin doses could reduce H2O2-induced damage. Results H 2 O 2 (0.3-0.9 mM) was dose-dependent and gossypin (5-200 µg/mL) inhibited proliferation to L929 cells after 75 µg/mL. 25 and 50 µg/mL doses of Gossypin protected L929 …
